CQ627 has a number of limitations for its use as a probe:
1. It has been reported to have a DC50 of 410 nM, which is at least one order of magnitude higher than ideal for its use as a probe.
2. Also, the relatively minor increase in the speed of degradation over the natural degradation of RIOK1 limits its usefulness.
3. The dependence on RNF126 has only been inferred by competition experiments with binders to RNF126, but not confirmed by knock-out or knock-down of RNF126.
4. Reports of a trimeric complex of RNF126, CQ627 and RIOK1 to confirm the claim that degradation is via RNF126 are lacking.
5. There are no proteomic studies reported, thus the selectivity of the probe is unknown.
